Results from people, animals, and cells

What Did AOD-9604 Research Find?

The mouse findings were hopeful. The study in people was not.

Which result matters most for people?

AOD-9604 changed how fat was made and burned in animals and cells. A beta-3 switch on mouse fat cells helped with that work. Those findings justified a human test without showing what would happen to people.

The human answer was much less hopeful. About 900 adults with obesity joined the full set of studies. In the main later trial, the placebo group lost just as much weight. As you read, note whether a result came from people, animals, or cells.

What did the AOD9604 peptide do to fat cells?

The AOD9604 peptide slowed new fat-making in lab work. One older test used growth hormone and a related end piece numbered 172–191. They touched the outer wall of fat and liver cells. That contact slowed a helper used to make fat [4].

Later studies tied this fat work to the short AOD9604 part [3]. A second finding came from obese mice. AOD-9604 increased a beta-3 switch on fat cells, which helps burn stored fat [1]. The amount moved closer to the level found in lean mice.

The compound avoids the cell doorway used by full growth hormone and leaves IGF-1, a growth-related blood number, unchanged [3]. This leaves out some of the hormone's wider work. An older rat-cell study also found a change in how sugar entered fat cells [8]. None of these lab results proves that a person will lose weight.

What did mice missing the fat-cell switch show?

One study compared obese mice with and without a working beta-3 switch, which fat cells use when burning stored fat. After fourteen days, growth hormone and AOD-9604 lowered weight and fat. The treatment also increased that switch [1]. Researchers then watched what happened in mice without it.

Those mice did not keep the longer weight and fat change [1]. They still had a brief rise in energy use and fat burning. The longer result therefore depended on the beta-3 switch. The study did not find the cause of the brief result.

A second paper also found more fat burning and weight loss in obese mice [2]. The two mouse findings agreed. Still, mice can't tell you whether a person will get the same result. That question required a study in people.

Did AOD9604 weight loss happen in people?

The AOD9604 weight loss studies did not show a clear gain for people. About 900 adults with obesity joined roughly six trials run by Metabolic Pharmaceuticals. They took AOD-9604 pills ranging from 0.25 mg to 54 mg each day. The studies lasted from 7 days to 24 weeks [5].

Reported problems were much the same with AOD-9604 and placebo [5]. In the main later trial, both groups lost about the same amount of weight. Work on the obesity drug stopped around 2007 [14]. A review later listed it among drugs that looked hopeful but never won approval [10].

The animal findings did not hold up in people. That is the strongest fact in this group of studies. Calling the compound a proven fat burner goes beyond the human results. The paper list lets you check each source.

Did the treatment help damaged rabbit knees?

A separate study looked at knee cartilage, the smooth covering on a joint. Researchers gave 32 New Zealand white rabbits arthritis in one knee. Each week, a knee received 0.25 mg AOD-9604, with or without 6 mg of hyaluronic acid, a slippery substance found in joints. After 4–7 weeks, treated knees had lower damage scores than knees given salt water [7].

This hopeful result came from rabbits and lab-grown cells. No published human arthritis trial has tested AOD-9604 itself. A separate rat study tested a related hormone piece and found a change in fat use [9]. Neither animal study answers whether AOD-9604 helps a person's knee.

How soon did the compound break apart in blood?

A pig study found that whole AOD-9604 stayed in blood for about 3 minutes after a vein shot. The body then removed its protein parts one at a time [6]. Other work showed that the body could take it in as a pill. Researchers therefore used tablets in the studies of people.

Special sport tests can find AOD-9604 in blood or urine [11][13]. That matters because sport rules ban the compound. Health officials have also found it in unapproved products taken from sellers [12]. Those products lacked the checks used in a study, so you can't trust what a vial contains.
